The Center for Cellular and Molecular Engineering (CCME) of the University of Pittsburgh is seeking a Research Technician. As a Research Technician in this lab, you will be a part of an exciting state-of-art internationally recognized tissue and cellular engineering research program in the area of musculoskeletal research, with a specific focus on cartilage and bone. The position is supported by a funded project and the Research Technician will work under the direct supervision of the senior scientists on the project. The candidate will manage cell and tissue cultures and assess cell proliferation, differentiation and metabolism, using RT_PCR, biochemistry, histology, immunohistochemistry and immunofluorescence (from sample preparation, to sectioning, to staining), etc. A major focus of the research will be on development of 3D microphysiological constructs maintained in unique bioreactor systems and scaffolds developed in the lab. The Research Technician is expected to maintain accurate records of laboratory procedures and results. Operating and maintaining sensitive laboratory equipment is also part of the job. The prompt communication of results, issues, and needs with team members is necessary. The candidate must have a good knowledge of laboratory skills and a high level of self-sufficiency. The capacity to work independently (including the design and execution of experiments) with some direction from senior scientists is required. Experience in literature search, identification of existing protocols and establishment/adaptation of protocol for specific cell culture conditions is expected as well as modifying standard procedures and methods to address the specific conditions of this project. Experience with mammalian cells culture is required. Experience in using optical microscopes in bright field mode and in fluorescence mode is required. Experience in either RT PCR or tissue histology/immunohistochemistry is required. This project requires the use of biochemical, histo/immune-histochemical and molecular techniques including, but not limited to, MTT, DMMB, alkaline phosphatase, TRAP, and hydroxyproline assays, DNA/RNA preparation and handling, RT PCR, western blotting and ELISAs, microscopy, basic histological staining, and immunohistochemistry (including immunofluorescence). Hence, the capacity to perform laboratory procedures and tests is expected. Capacity of using the Office suite, including Excel, is required, as is the capacity to perform the basic calculations for dilutions, standard curves, and quantification of data obtained by assays such as RT PCR or colorimetric assays. Multi-tasking several experiments simultaneously and efficient time management is necessary. Many of the responsibilities described above will be self-initiated and require a high level of vigilance. Meticulous work habits, dependability, and good communication skills are required. Two year's experience of wet lab work are expected. Bachelor's Degree or equivalent in biology, chemistry, molecular biology, biochemistry, bioengineering, or related field is required. Experience in mammalian cell culture is required. . Experience in optical microscopes (bright field/fluorescence) is required. Experience in either RT PCR or tissue histology/immunohistochemistry is required, Emphasis in musculoskeletal tissues/cells is an asset. Three years of experience is preferred.
All regulatory compliance modules regarding laboratory safety, EHS, etc. before beginning in the lab (will be provided locally).
